Can't dial some conferences

I inherited a FreePBX with several conferences set up. Most of them can be dialed internally by simply dialing the conference number (e.g. 81234). However, the “main” conference line is set up as a destination for a DID and can only be called via the DID. Trying to dial the conference directly doesn’t work (“Your call cannot be completed as dialed.”).

As far as I can tell, this conference is not configured any differently from the ones that work. What could I be missing?

I would delete it. I would recreate it and make sure I can get to it internally. If everything looks good then go to your inbound route point it to your new conference. Everything should be good.
